Imagine you are going to email a professor in the field of physics in order to get a research position. In text #1, you can see a sample text for such a topic:
Subject: Research position – Pulsed, low-temperature supersonically cooled OH molecules
Dear Dr. Smith,
I am applying to North Carolina State University for fall 2011 for MS in Physics. I have read few of your research publications on pulsed, low-temperature supersonically cooled OH molecules. I have worked on a similar research project in my undergraduate involving OH molecules at hyper-state and below sub-degree level. I am interested in your research area and would like to know about research opportunities to work with you.
Looking forward for your reply.

Arash Rad

Sometimes you need to have a letter of recommendation from your supervisor so that you can present it to your future employer. In text number #2, you can see a sample text of this.
Subject: Behrouz Javan Reference Request
Dear Dr. Jones,
I am writing to you to request that you provide a reference for me as I begin my job search. As you know, I will be completing my graduate studies this spring, and have found several exciting opportunities that I am exploring.
As my undergraduate thesis advisor and mentor, I believe that a reference from you would provide a potential employer with information to recommend me as a school counselor.
If you need any additional information, please contact me via email or phone.
Thank you for your consideration and support.
Sincerely,
Behrouz Javan
