A) We mix 100u of 2ME buffer with 100u of anti-D and incubate for one hour at 37 degrees.
b) The product of step A is mixed with 6200 Landa physiological serum and 3200 Landa O positive suspension washed three times (we use 150 Landa RBC with 10 cc of physiological serum to prepare the suspension) and incubate for 30 minutes at 37 degrees.

Important note: Centrifuge the product of step C before storing it in the refrigerator. Sometimes, due to the high titer of anti-D, the product of step C will agglutination by itself after centrifugation (without adding anti-human globulin). To solve this problem, you must prepare serial dilutions of the product of step A and enter each dilution separately into the next steps. Usually, in a 1:4 dilution of the product of step A, the final mixture (product of step C) will not have any spontaneous agglutination either macroscopically or microscopically. This mixture is stored in the refrigerator as a tuberculosis check and usually lasts up to three weeks.
– To control the anti-human globulin, mix 100 L of Chek Sel with 200 L of anti-human in one tube, and as a control to ensure the absence of spontaneous agglutination, mix 100 L of Chek Sel with 200 L of physiological serum and according to the last step Indirect Coombs, after staying at room temperature for 5 minutes, we centrifuge. The anti-human tube should have macroscopic or at least microscopic agglutination and the physiological serum tube should not have any agglutination.
